Kim, Nari, Jin Han, and Euiyong Kim. Effects of prostaglandin F2 on membrane currents in rabbit middle cerebral arterial smooth muscle cells. Am J Physiol Heart Circ Physiol 284: H1018–H1027, 2003; 10.1152/ajpheart. 01022.2001.—Although PGF2 affects contractility of vascular smooth muscles, no studies to date have addressed the electrophysiological mechanism of this effect. The purpose of our investigation was to examine the direct effects of PGF2 on membrane potentials, Ca2 -activated K (KCa) channels, delayed rectifier K (KV) channels, and L-type Ca2 channels with the patch-clamp technique in single rabbit middle cerebral arterial smooth muscle cells (SMCs). PGF2 significantly hyperpolarized membrane potentials and increased the amplitudes of total K currents. PGF2 increased open-state probability but had little effect on the open and closed kinetics of KCa channels. PGF2 increased the amplitudes of KV currents with a leftward shift of the activation and inactivation curves and a decrease in the activation time constant. PGF2 decreased the amplitudes of L-type Ca2 currents without any significant change in threshold or apparent reversal potentials. This study provides the first finding that the direct effects of PGF2 on middle cerebral arterial SMCs, at least in part, could attenuate vasoconstriction.

Effects of prostaglandin F2alpha on membrane currents in rabbit middle cerebral arterial smooth muscle cells.

Although PGF(2alpha) affects contractility of vascular smooth muscles, no studies to date have addressed the electrophysiological mechanism of this effect. The purpose of our investigation was to examine the direct effects of PGF(2alpha) on membrane potentials, Ca(2+)-activated K(+) (K(Ca)) channels, delayed rectifier K(+) (K(V)) channels, and L-type Ca(2+) channels with the patch-clamp techniq...

TMEM16A channels generate Ca²⁺-activated Cl⁻ currents in cerebral artery smooth muscle cells.

Transmembrane protein (TMEM)16A channels are recently discovered membrane proteins that display electrophysiological properties similar to classic Ca(2+)-activated Cl(-) (Cl(Ca)) channels in native cells. The molecular identity of proteins that generate Cl(Ca) currents in smooth muscle cells (SMCs) of resistance-size arteries is unclear. Similarly, whether cerebral artery SMCs generate Cl(Ca) c...

[Roles of prostaglandin on rabbit vesicourethral smooth muscle contraction].

The effects of prostaglandin (PG) E1, E2 and F2 alpha on isolated smooth muscles of rabbit bladder and urethra were studied by in vitro techniques for recording contractile activities. To examine the mechanism of PGs' effect, intracellular cyclic AMP content was also measured by radioimmunoassay. Spontaneous contractile force of muscle strips isolated from rabbit urinary bladder dome and base w...

Activation of inward rectifier K+ channels by hypoxia in rabbit coronary arterial smooth muscle cells.

We examined the effects of acute hypoxia on Ba2+-sensitive inward rectifier K+ (K(IR)) current in rabbit coronary arterial smooth muscle cells. The amplitudes of K(IR) current was definitely higher in the cells from small-diameter (<100 microm) coronary arterial smooth muscle cells (SCASMC, -12.8 +/- 1.3 pA/pF at -140 mV) than those in large-diameter coronary arterial smooth muscle cells (>200 ...
